Dr. Shamsheer Vayalil was born on January 11, 1977, in Kozhikode, Kerala, India. Born into a business family, his father is Hashim Pokkinari, and his mother is Mariyam Barakkool. Dr. Shamsheer Vayalil is married to Dr. Shabeena Yusuffali, the eldest daughter of billionaire businessman M. A. Yusuff Ali. He has four children.
Dr. Shamsheer Vayalil earned his Bachelor of Medicine and Bachelor of Surgery (MBBS) degree from Kasturba Medical College, Manipal. He later pursued a Doctor of Medicine (MD) in Radiology from Sri Ramachandra Medical College & Research Institute, Chennai.
